Título : Elaboración de películas con actividad antioxidante a base de alginato y polifenoles extraídos de las macroalgas pardas de Macrocystis pyrifera y Lessonia trabeculata

Resumen : Las macroalgas han sido utilizadas desde la antigüedad como fuente de alimentos por su alto valor nutricional. Actualmente, en las industrias se emplean las macroalgas pardas para la obtención de alginato, el cual por su variabilidad es empleado en la industria cosmética, farmacéutica y alimentaria como agentes espesantes, moldeadores y gelificantes según sea su calidad. El Perú cuenta con una diversidad de macroalgas, las cuales contienen diferentes polisacáridos y metabolitos bioactivos que pueden obtenerse en una extracción por fases, siendo estos el alginato y los compuestos polifenólicos, de este modo se busca aprovechar y maximizar el proceso de extracción. La característica gelificante del alginato lo hace factible para la elaboración de películas; adicionalmente, el empleo de aditivos naturales bioactivos confieren a las películas la acción antioxidante, antimicrobiana entre otros. Por tal motivo, esta investigación se enfocó en aprovechar el proceso de extracción a fin de obtener las materias primas para la elaboración de películas bioactivas. Este trabajo muestra los análisis comparativos del contenido de fenoles totales y la actividad antioxidante de dos macroalgas pardas en distintos medios de extracción; con valores de 94.13 y 187.70 mg EAG/100g MS para los extractos hidroalcohólicos de las hojas de la M. pyrifera y L. trabeculata respectivamente. Además, el potencial antioxidante de los extractos frente al DPPH fue del 81.74 % y 84.45 % de la M. pyrifera y L. trabeculata respectivamente. A partir de las macroalgas pardas se obtuvo también el alginato de sodio con rendimientos de extracción del 17.99 % de la M. pyrifera y 26.89 % de la L. trabeculata. La caracterización de los alginatos obtenidos se realizó mediante su análisis por FTIR, TGA, DRX, RMN1H, RMN13C en estado sólido y se determinó la masa molar promedio por ensayos de viscosimetría capilar. A partir de los alginatos y de los extractos hidroalcohólicos extraídos de las macroalgas pardas se elaboraron películas bioactivas, los cuales fueron analizados por sus propiedades físicas (solubilidad, humedad, espesor), caracterizadas por FTIR, TGA, DRX, opacidad, FESEM y se determinó la capacidad antioxidante de cada película, siendo mayor la acción en películas cargadas con extractos de L. trabeculata; asimismo, las películas evidenciaron un incremento de la opacidad al agregar los extractos. En adición, el análisis preliminar de las películas bioactivas frente a paltas Hass evidenció una reducción del pardeamiento y deterioro del fruto hasta 21 días del análisis.
Macroalgae have been used since ancient times as a source of food for their high nutritional value. Currently, brown macroalgae are used in industries to obtain alginate, which due to its variability is used in the cosmetic, pharmaceutical and food industries as thickening, molding and gelling agents depending on their quality. Peru has a diversity of macroalgae, which have different contents of polysaccharides and biactive metabolites that can be obtained in a phased extraction, these being alginate and polyphenolic compounds, in this way we seek to take advantage and maximize the ectraction process. The gelling characteristic of alginate makes it feasible for making films; additionally, the use of bioactive additives gives the films antioxidant and antimicrobial action, among others. For this reason, this research focused on taking advantage of the extraction process in order to obtain raw materials for the production of bioactive films. This work shows the comparative analyzes of the total phenolic content and antioxidant activity of two brown macroalgae in different extraction solvents; with TPC values of 94.13 and 187.70 mg GAE/100g dw for the hydroalcoholic extracts of the leaves of M. pyrifera and L. trabeculata respectively. In addition, the antioxidant potential of the extracts against DPPH was 81.74 % and 84.45 % of M. pyrifera and L. trabeculata respectively. From brown macroalgae, sodium alginate was also obtained with extraction yields of 17.99 for M. pyrifera and 26.89 for L. trabeculata. The characterization of the alginate was determined by FTIR, TGA, XRD, 1H- NMR, 13C-NMR in solid state and the average molar mass was determined by capillary viscometry tests. From the alginates and the hydroalcoholic extracts obtained from the brown macroalgae, bioactive films were prepared, which were analyzed for their physical properties (solubility, humidity and thickness), characterized by FTIR, TGA, XRD, opacity, FESEM and the antioxidant capacity of each film was determined, with an inhibition percentage between 40 – 50 %; likewise, the films showed an increase in opacity when the extracts were added. In addition, the preliminary analysis of the bioactive films agasinst Hass avocados indicated a reduction in browning and deterioration of the fruit up to 21 days after the analysis.
